I imagine the best way of going about this would be to first create an archetype of absolute normality, with specific characteristics that can be measured objectively (or, as objectively as possible). To decide an individual's NT levels, their characteristics could be compared to the archetype, and if they stray too far from this archetype, then they would be considered to be non-NT.
I should say that this would not include the use of brain scans or other advanced technologies, as we do not have the technologies for that. But rather, hypothetically at least (idk if anyone could be bothered to do such a thing) we could create some sort of a system for this purpose.

The archetypal traits of absolute normality could perhaps include such things as a positive attitude to socialising... but this would have to be refined. But a problem is that this archetype is malleable to forever changing social attitudes.
*Sighs loudly* I bet something like this had already been created. I'm just wasting my time and I still feel drunk from last night. Ugh. I might as well post anyway since I've typed this all out.
